A7 Agar Base (Shepard’s Differential Agar Base) is recommended for cultivation and differentiation of Ureaplasma urealyticum from urine based on its ability to produce ammonia from urea. It is also used for the cultivation of other Ureaplasma species. Ureaplasma possess the enzyme urease and hence breakdown the urea to ammonia. Ureaplasma urealyticum produces ammonia hence appear as golden to dark brown coloured colonies.
