Intended Use
Recommended for the determination of total yeast and mould count in food, dairy, water and waste water samples.

Directions
Open the pouch aseptically. Open the lid and add the required dilution of 0.5 ml to 1 ml. The solution will be evenly absorbed. Close the lid. Press the sides of the lid to ensure that it is fixed in the grooves. Allow 5-10 minutes for even absorption. Incubate the plates in horizontal position at specified temperature and period. After incubation, count the number of colonies which have appeared on the surface of the medium.
Principle and Interpretation
HiPetriSlim™™ Total Yeast and Mould Count are recommended to check critical control points in protected areas while establishing or revising microbiological surface monitoring techniques. The medium is highly nutritious and provides amino acids and long chain peptides for the growth of microorganisms. This medium is used for stimulating sporulation, for maintaining stock cultures of certain dermatophytes and for differentiation of typical varieties of dermatophytes on the basis of pigment production. It is also recommended for growth of fungi. The medium promote luxuriant fungal growth. The pH of the medium is adjusted to 3.5, that inhibits the bacterial growth.

Specimen Collection and Handling

After use, contaminated materials should be autoclave at 121°C for 15 minutes before discarding.
Warning and Precautions
Read the label before opening the container. Wear protective gloves/protective clothing/eye protection/ face protection. Follow good microbiological lab practices while handling specimens and culture. Standard precautions as per established guidelines should be followed while handling specimens. Safety guidelines may be referred in individual safety data sheets.
Limitations
- Do not use diluents containing citrate, bisulfite or thiosulfate; they can inhibit growth.
Performance and Evaluation
Performance of the medium is expected when used as per the direction on the label within the expiry period when stored at recommended temperature.

Sterility Check: Passes release criteria.
Cultural Response: Cultural characteristics observed after an incubation at 22 - 28°C for 48 - 72 hours.

Storage and Shelf Life
Store between 2-8°C. Use before expiry date on the label. Product performance is best if used within stated expiry period.
Disposal
User must ensure safe disposal by autoclaving and/or incineration of used or unusable preparations of this product. Follow established laboratory procedures in disposing of infectious materials and material that comes into contact with sample must be decontaminated and disposed of in accordance with current laboratory techniques (1,2).
