Formula adjusted, standardized to suit performance parameters.

Directions

Suspend 30.0 grams in 1000 ml purified/distilled water. Heat if necessary to dissolve completely. Mix well and dispense as desired. Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes.

If any fibres are observed, filter the solution through a 0.22 µm filter to eliminate them.

Principle and Interpretation

Soyabean Casein Digest Medium is recommended by various pharmacopeias for sterility and microbial limit testing. It is highly nutritious, supporting a wide range of organisms. Tryptone and soya peptone supply nitrogen, amino acids, and peptides; glucose provides carbohydrate; phosphate buffers the medium; sodium chloride maintains osmotic balance.

Pharmaceutical samples; clinical samples such as urine, pus, and wound exudates.

For clinical samples, follow standard handling techniques per established guidelines. For pharmaceutical samples, adhere to pharmacopeial procedures. Contaminated materials must be sterilized by autoclaving before disposal.

Limitations

  • Further biochemical characterization of pure colonies is required for identification.

Appearance: Cream to yellow homogeneous free flowing powder.

Colour and Clarity: Light yellow clear solution without precipitate.

Reaction: pH 7.3 ± 0.2 (3% w/v aqueous solution at 25°C after sterilization).

Stability Test: Clear yellow solution stable for 7 days at room temperature.

Storage and Shelf Life

Store between 10–30°C in tightly closed container. Prepared medium: 15–30°C. Protect from moisture and ignition sources. Seal tightly after use. Best used within stated expiry period.

Disposal

Dispose by autoclaving and/or incineration of used media. Follow biosafety and laboratory protocols for infectious waste handling.
