
Soyabean Casein Digest Medium (Tryptone Soya Broth)
Intended Use
Recommended as a general purpose medium used for cultivation of a wide variety of microorganisms and recommended for sterility testing of moulds and lower bacteria.

Directions
Suspend 30.0 grams in 1000 ml purified/distilled water. Heat if necessary to dissolve completely. Mix well and dispense as desired. Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes.
If any fibres are observed, filter the solution through a 0.22 µm filter to eliminate them.
Principle and Interpretation
Soyabean Casein Digest Medium is recommended by various pharmacopeias for sterility and microbial limit testing. It is highly nutritious, supporting a wide range of organisms. Tryptone and soya peptone supply nitrogen, amino acids, and peptides; glucose provides carbohydrate; phosphate buffers the medium; sodium chloride maintains osmotic balance.

Limitations
- Further biochemical characterization of pure colonies is required for identification.
- This general-purpose medium may not support growth of fastidious organisms.

Quality Control
Appearance: Cream to yellow homogeneous free flowing powder.
Colour and Clarity: Light yellow clear solution without precipitate.
Reaction: pH 7.3 ± 0.2 (3% w/v aqueous solution at 25°C after sterilization).
Stability Test: Clear yellow solution stable for 7 days at room temperature.

Storage and Shelf Life
Store between 10–30°C in tightly closed container. Prepared medium: 15–30°C. Protect from moisture and ignition sources. Seal tightly after use. Best used within stated expiry period.
Disposal
Dispose by autoclaving and/or incineration of used media. Follow biosafety and laboratory protocols for infectious waste handling.
