Sulphorhodamine B sodium salt is a fluorescent dye used to determine the amount of cellular proteins in cultured cells and in cytotoxicity assays based on fluorescence or colorimetric output. The dye has maximal absorbance at 565 nm light and maximal fluorescence emission at 586 nm light.
It binds stoichiometrically to proteins under mild acidic conditions and then can be removed using basic conditions. Thus, the amount of bound dye can be used as an approximate for cell mass, which can then be extrapolated to measure cell proliferation.
