Nutrient HiVeg Agar Medias are recommended as general purpose culture media for microbiological analysis of water and cultivation of bacteria requiring slightly acidic pH.

Directions

Suspend 40 grams of MV1269 or 31 grams of MV090 in 1000 ml distilled water. Heat to boiling to dissolve the medium completely. Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes.

Principle and Interpretation

These media are prepared by completely replacing animal based peptones with vegetable peptones, which makes the media free of BSE/TSE risks. HiVeg peptone and HiVeg extract are used in these medias as veg substitute in place of Peptic digest of animal tissue and Beef extract respectively.

Nutrient HiVeg Medias are the modification of Nutrient Media which are general purpose media used for the examination of water and dairy products according to Standard Methods for the Examination of Water and Wastewater (1) and Dairy Products (2). Nutrient HiVeg Agar No. 2 like the conventional Nutrient Agar No.2 can be used for the Microbiological analysis of water as per Czech Standards. It can also be used for sterility testing of aerobes and maintenance of subcultures (3). Nutrient HiVeg Agar with 0.8% NaCl and pH 6.0 which is a modification of Nutrient Agar w/ 0.8% NaCl and pH 6.0 recommended by APHA for the growth of microorganisms requiring slightly acidic conditions.

HiVeg peptone and HiVeg extract provides the necessary nitrogen compounds, carbon, vitamins and also some trace ingredients necessary for the growth of bacteria. Sodium chloride maintains osmotic equilibrium of the medium.

Quality Control

Appearance of powder: Light yellow to yellow coloured may have slightly greenish tinge, homogeneous, free flowing powder.

Gelling: Firm, comparable with 1.5% Agar gel of MV1269 or MV090.

Colour and Clarity: Light yellow to amber coloured, clear to slightly opalescent gel forms in petri plates.

Reaction

Reaction of 4.0% w/v aqueous solution of MV1269 is pH 7.2 ± 0.2 at 25°C. Reaction of 3.1% w/v aqueous solution of MV090 is pH 6.0 ± 0.2 at 25°C.

Cultural Response

Cultural characteristics observed after an incubation at 35 - 37°C for 18-24 hours of MV1269.
