Blocking Buffer (10% BSA in 1X TBS)

Blocking Buffer (10% BSA in 1XTBS) is made of high-quality BSA that is useful for saturating excess protein-binding sites on membranes and microplates in immunoassays. The Blocking Buffer (10%) should be diluted up to 1% working concentration prior to use with 1X Tris-buffered saline. A good blocking buffer for a specific experiment will bind to all potential sites of nonspecific interaction, eliminating background without altering or obscuring the epitope for antibody binding. In case of immunoblotting/staining of phosphorylated protein TBS is recommended instead of PBS, because phosphate in PBS may interfere with immunoreaction between phospho protein and phospho specific Antibodies. Blocking Buffer (10% BSA in 1X TBS) is a Tris based formulation designed for assays where phosphate ions might interfere with alkaline phosphatase or other reagents. This blocking buffer supports background reduction in molecular immunology diagnostic research and Western blotting.
