Introduction

X-Press Blue™ Protein Stain (SafeBlue Protein Stain) is CoomassieⓇ G-250 based rapid, ultra-sensitive ready-to-use staining solution used in visualizing protein bands on SDS-PAGE. Unlike other gel staining systems no destaining is required while using X-Press Blue™ Protein Stain (SafeBlue Protein Stain).

Description

X-Press Blue™ Protein Stain (SafeBlue Protein Stain) is a ready-to-use solution containing Coomassie Brilliant Blue G-250 at a low pH and can detect 200 ng of protein. This protein staining reagent has higher sensitivity and gives lower background compared to the conventional coomassie stains. This ready to use reagent develops visible protein bands directly in the staining tray. Thus, usage of this solution saves a lot of time required for solution preparation. After staining, an optional water-wash step further enhances staining sensitivity and yields a very low background. With this reagent there is no need for multistep destaining procedures. This reagent stains only proteins and leaves the gel background relatively unstained so that the protein bands are visualized directly during the staining process. This results in the saving of time as the staining can be stopped when the protein bands have been properly stained. Although a conventional destaining step with methanol and acetic acid is not required as with other Coomassie-based stains, rinsing the stained gel with water enhances the sensitivity.

Composition

X-Press Blue™ Protein Stain (SafeBlue Protein Stain) solution contains Coomassie G-250 in a proprietary solution.

Procedure

1. After performing SDS-PAGE rinse the mini-gel with 100 ml of distilled water 3 times for 5 minutes with gentle shaking. Discard the water after each wash.
2. Stain the gel with enough SafeBlue Protein Stain (approx. 20 ml) to submerge the entire gel.
3. Stain for an hour at room temperature with gentle shaking. Discard the staining solution.
NOTE: SafeBlue Protein Stain is for one time usage only. Do not reuse it.
4. Wash the gel with 100 ml of distilled water for a minimum of one hour. Longer wash times will increase band intensity and reduce background.
6. A second water wash for one hour to overnight is required for a clearer background.

Storage conditions

X-Press Blue™ Protein Stain (SafeBlue Protein Stain) has to be stored at room temperature (15-25°C). Under recommended condition, the reagent is stable for 12 months.

Warning and Precautions

Not for Medicinal Use. Read the SDS carefully before beginning the protocol. Wear protective gloves/protective clothing/eye protection/face protection. Follow good clinical laboratory practices while handling clinical samples. Standard precautions should be followed as per established guidelines. Performance and Evaluation

Performance of the solution is expected when the solution is stored at recommended temperature and within the expiry period.
