Denaturation Solution is a ready to use buffer solution consists of Sodium chloride and strong base Sodium hydroxide which is especially prepared for use in Southern Blotting and in other in situ hybridization procedures.
Denaturation Solution denatures the double-stranded DNA and as a result DNA can be efficiently transferred to nylon or nitrocellulose membrane and subsequently hybridized to a labeled probe during neutral Southern blotting procedure. When denaturation solution is used in conjunction with depurination solution, the phosphodiester backbone at the sites of depurination is hydrolysed which allows a very efficient transfer of large DNA to the membrane.
