Motility Nitrate Medium, Buffered is recommended for isolation and detection of Clostridium perfringens on the basis of motility and nitrate test. Peptone and HM peptone B supply amino acids and other complex nitrogenous substances. Medium being semisolid gel is used to demonstrate motility of the organism. Growth of motile organisms extends away from the line of inoculation along with turbidity. In the nitrate reduction test, on addition of Nitrate reagents a pink to red color develops if nitrite is present. Clostridium perfringens is positive for nitrate reduction and nonmotile. Clostridium absonum is weakly motile and shows weak or negative nitrate reduction.
