HiPurA® Competent Cells are specially prepared chemically competent DH10B cells which makes cloning experiments easier. Competent cells are ready to use bacterial cells that contain an altered cell membrane through which a foreign DNA can pass through very easily. HiPurA® Competent Cells are prepared by following a modified protocol of Hanahan (1983, J.Mol.Biol 166:557) and are recommended for cloning of higher molecular weight recombinant plasmids (Greater than 10kb-15kb).
HiPurA® Competent Cells are prepared by following a modified protocol of Hanahan (1983, J.Mol.Biol 166:557) and are recommended for cloning of higher molecular weight recombinant plasmids (Greater than 10kb-15kb). DH10B is the most frequently used E.coli strain for routine cloning applications through blue/white screening of colonies on plates containing X-Gal and IPTG as the ? 80lacZ?M15 marker provides ?complementation of the ?-galactosidase gene. A stock pUC19 solution is provided as a control to determine the transformation efficiency. To obtain maximum transformation efficiency, the experimental DNA must be free of phenol, ethanol, protein and detergents.
