Intended Use

Directions

This product is available in multiple pack sizes of 25 and 50 bottles containing 10 ml of sterile medium in each bottle.

  1. The bottles when supplied are intact. Ensure that all bottles are in upright position and there is no leakage or any manufacturing defect or contamination.
  2. User may remove the desired number of bottles from the box as per their requirement.
  3. It should be handled by trained person wearing appropriate personal protective equipment (PPE) and sterile gloves.
  4. Place the bottles on sterile surfaces such as laminar air flow or sterile working bench.
  5. Label them accordingly.
  6. Disinfect the outer surface of cap or closures with suitable disinfectant example 70% IPA.
  7. Observe aseptic techniques and standard microbiological methods while processing and inoculation of samples or cultures into this media.
  8. On completion of inoculation tighten the cap and close it carefully.
  9. Incubate at specified temperature and time or as desired.
  10. Follow good lab practices for procedures and disposal.

Principle And Interpretation

Cooked Meat Medium w/Glucose, Hemin and Vitamin K is a modification of the Cooked Meat Medium originally developed by Robertson (1) for cultivation of anaerobes isolated from wounds. Moore et.al (2) have recommended this modified medium for subculturing of anaerobic isolates to be examined by gas liquid chromatography.

The medium contains HM granules B, which provides amino acids and nutrients. It also contains glutathione, a reducing substance which permits the growth of obligate anaerobes. The sulphydryl groups which impart reducing effect are more available in denatured protein and hence the cooked meat is added in the medium. The added supplements glucose, yeast extract, hemin and vitamin K act as growth enhancers for anaerobic microorganisms. The growth in this medium is indicated by the turbidity or bubble formation by some organisms. Blackening and disintegration of the peptone indicate proteolysis.

Specimen Collection and Handling

For clinical samples follow appropriate techniques for handling specimens as per established guidelines (3,4). After use, contaminated materials must be sterilized by autoclaving before discarding.

Limitations :

  1. Individual organisms differ in their growth requirement and may show variable growth patterns on the medium.
  2. Each lot of the medium has been tested for the organisms specified on the COA. It is recommended to users to validate the medium for any specific microorganism other than mentioned in the COA based on the user's unique requirement.
  3. Biochemical characterization is carried out from pure isolates for complete identification.
  4. Recommended for Pediatric use only.

Performance and Evaluation

Performance of the medium is expected when used as per the direction on the label within the expiry period when stored at recommended temperature.

Store between 2-8°C. Use before expiry date on the label. Product performance is best if used within stated expiry period.

Disposal

User must ensure safe disposal by autoclaving and/or incineration of used or unusable preparations of this product. Follow established laboratory procedures in disposing of infectious materials and material that comes into contact with clinical sample must be decontaminated and disposed of in accordance with current laboratory techniques (3,4).
