In cell culture applications, Dispase® is used as a cell dissociation reagent for many cell types. Dispase® retains cell-cell contact and permits cell differentiation. Dispase® is isolated from bacteria hence does not carry the risk of contamination with Mycoplasma or other adventitious agent. Dispase® does not cleave intracellular linkages but cleaves only linkages between substrate and cell sheet, hence the cells can be recovered in form of an entire cell sheet and can be seeded. Dispase® is active even in presence of serum and can be added in suspension cultures to avoid unwanted cell clumping. Cell survival upon dissociation is higher compared to trypsin.
