Directions
Streak the test inoculum aseptically into the slant and incubate at appropriate conditions. Incubate the slants at 30-35°C for 18-24 hours.
Principle And Interpretation
BHI Agar is highly nutritious and can support luxuriant growth of wide variety of microorganisms. It can be further enriched by the addition of blood or rendered selective by adding different antibiotics (8, 2). It is a general purpose medium used for primary isolation of aerobic bacteria from clinical specimens. Addition of 50 mg/l chloramphenicol or 40mg/l streptomycin or a mixture of 50mg/l gentamicin and 50mg/l chloramphenicol along with 5-10% sterile defibrinated blood is often recommended for inhibition of bacteria and isolation of pathogenic systemic fungi. (6). Some fungi may be inhibited on this medium with 10% sheep blood, gentamicin and chloramphenicol (3,7,1).
Proteose peptone and infusions used in the media serves as sources of carbon, nitrogen, vitamins, amino acids, along with essential growth factors. Dextrose is the energy source. Sodium chloride maintains the osmotic equilibrium of the medium while disodium phosphate buffers the medium. Defibrinated sheep blood added to the basal medium provides essential growth factors for the more fastidious fungal organisms.

Specimen Collection and Handling
For clinical samples follow appropriate techniques for handling specimens as per established guidelines (4,5).
After use, contaminated materials must be sterilized by autoclaving before discarding.
Warning and Precautions
In Vitro diagnostic use. Read the label before opening the container. Wear protective gloves/protective clothing/ eye protection/face protection. Follow good microbiological lab practices while handling specimens and culture. Standard precautions as per established guidelines should be followed while handling clinical specimens. Safety guidelines may be referred in individual safety data sheets.
Limitations
- As organisms differ in their nutritional requirements, some fastidious organisms may be inhibited or may show poor growth.
- Further biochemical tests must be carried out for complete identification.
Performance and Evaluation
Performace of the medium is expected when used as per the direction on the label within the expiry period when stored at recommended temperature.

Storage and Shelf Life
On receipt store between 2-8°C Use before expiry date on the label. Product performance is best if used within stated expiry period.
Disposal
User must ensure safe disposal by autoclaving and/or incineration of used or unusable preparations of this product. Follow established laboratory procedures in disposing of infectious materials and material that comes into contact with clinical sample must be decontaminated and disposed of in accordance with current laboratory techniques (4,5).
