ISP HiVeg Medium No. 6 (Peptone Yeast Extract Iron HiVeg Agar) is recommended for the cultivation and maintenance of Streptomyces species.

Directions
Suspend 37.58 grams in 1000 ml distilled water. Heat to boiling to dissolve the medium completely. Dispense as desired and sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes. Cool to 45-50°C. Pour into sterile petriplates.
Principle and Interpretation
This medium is prepared by using HiVeg peptone and HiVeg peptone No.3 in place of Peptic digest of animal tissue and Proteose peptone making the medium free of BSE/TSE risks. ISP HiVeg Medium No. 6 (Peptone Yeast Extract Iron HiVeg Agar) is the modification of ISP Medium No. 6 (Peptone Yeast Extract Iron Agar) according to the International Streptomyces Project for the cultivation and maintenance of Streptomyces species (1, 2). HiVeg peptone, HiVeg peptone No.3 and yeast extract provide carbon, nitrogen, sulphur, vitamin B complex and other essential growth nutrients. Dipotassium hydrogen phosphate gives the medium good buffering capacity.

Quality Control
Appearance of powder
Yellow coloured, may have slightly greenish tinge, homogeneous, free flowing powder.
Gelling
Firm, comparable with 1.5% Agar gel.
Colour and Clarity
Yellow coloured, clear to slightly opalescent gel forms in petri plates.
Reaction
Reaction of 3.76% w/v aqueous solution is pH 6.7 ± 0.2 at 25°C.
Cultural Response
Cultural characteristics observed after an incubation at 25-30°C for 48 - 72 hours.
