Description of the product

2X Urea Denaturing Gel Loading Buffer is recommended for use with polyacrylamide urea gel (denaturing) agarose gel electrophoresis of nucleic acids. 2X Urea Denaturing Gel Loading Buffer is suitable for use in loading nucleic acid samples in polyacrylamide urea gel (denaturing) and non-denaturing agarose gel electrophoresis of nucleic acids.

Procedure

Sample preparation and Loading on Agarose Gels:

1. Add 25µl of ML176 to 25µl of sample and Mix properly.
2. Load 10µl of prepared sample in 1% AGE gel
3. Run the gel in 1X TAE/ TBE Buffer till the dye front reaches three-fourth of the gel.

NOTE: Prestaining of the gel with Ethidium bromide is recommended

Sample Preparation and Loading on Urea PAGE:

1. Add 25µl of ML176 to 25µl of sample and Mix properly.
2. Loading can be done with or without heating.
3. Heating can be done at 95°C for 5 minutes.
4. Load 10µl of prepared sample in Urea-PAGE gel
5. Run the gel in 1X TBE Buffer at 130V for 55mins.

NOTE: For Urea denaturing PAGE, post staining of the gel with Ethidium bromide is recommended.

Not for Medicinal Use. Read the SDS carefully before beginning the protocol. Wear protective gloves/protective clothing/eye protection/face protection. Follow good clinical laboratory practices while handling clinical samples. Standard precautions should be followed as per established guidelines. Performance of the solution is expected when the solution is stored at recommended temperature and within the expiry period.
