10X Citrate buffer, pH 6.1, is extensively used on formalin-fixed, paraffin-embedded tissue sections mounted on glass slides during immunostaining procedures. In this process tissue samples have to be fixed in an appropriate fixative for prevention of autolysis, preservation of tissue structures and immobilization of antigens. However, this results into structural alteration of antigens and subsequent masking of the reactive sites. 10X Citrate Buffer, after diluting to 1X, is used to recover antigens masked by fixation in cross-linking fixatives
