Directions

Suspend 2.0 grams in 1000 ml distilled water. Sterilize by autoclaving at 10 lbs pressure (115°C) for 20 minutes. Mix well and dispense as desired.

Principle and Interpretation

Synthetic Complete Supplement Mixture (SC) is used as an amino acid supplement for all strains of Saccharomyces cerevisiae. This yeast strain is called budding yeast and is extensively studied microorganism in molecular and cell biology.

Synthetic Complete Supplement Mixture (SC) is developed according to the formulae of Wickerham (1, 2) which supplies the essential nutritional elements for budding yeast cells. It is composed of all the amino acids required for the vigorous growth of Saccharomyces cerevisiae. It can be mixed with yeast nitrogen base (YNB), ammonium sulphate and an appropriate carbon source (glucose/galactose/raffinose) to produce a media suitable for the growth of budding yeast. It is also the basis for many “Drop-Out” formulations for the selections of auxotrophic requirements and transformants.

Quality Control

Appearance of Powder: White to cream coloured, homogeneous, free flowing powder.

Colour and Clarity: Colourless, clear solution without any precipitate.

Cultural Response: Cultural characteristics observed after an incubation at 25-30°C for 18 - 48 hours.
