Directions

Suspend 51.7 grams in 1000 ml distilled water. Sterilize by autoclaving at 10 lbs pressure (115°C) for 20 minutes. Mix well and dispense as desired.

Principle and Interpretation

DOBA Growth Medium with 2% Galactose and 1% Raffinose is used for the growth of Saccharomyces cerevisiae. This yeast strain is called budding yeast and is extensively studied microorganism in molecular and cell biology.

DOBA Growth Medium with 2% Galactose and 1% Raffinose is developed according to the formulae of Wickerham (1, 2) which supplies the required nutritional elements for budding yeast cells. This media is used for the classification of yeast strains based on the carbon and nitrogen requirements and it contains all the essential vitamins and inorganic salts except Ammonium sulphate which is required for the propagation of yeast cells. Furthermore, this media does not contain the amino acids namely, histidine, methionine, leucine, lysine and tryptophan. Galactose and raffinose serve as the carbon source for the cells.

Quality Control

Appearance of Powder: Light yellow coloured, homogeneous, free flowing powder.

Gelling: Firm, comparable with 1.5% Agar gel.

Colour and Clarity: Light yellow coloured, clear to slightly opalescent gel forms in Petri plates.

Cultural Response: Cultural characteristics observed after an incubation at 25-30°C for 18 - 48 hours.
