Directions
Suspend 36 grams in 1000 ml distilled water. Heat to boiling to dissolve the medium completely. Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes. Mix well and dispense as desired.
Principle and Interpretation
Sporulation Growth Agar is used for the growth and sporulation of Saccharomyces cerevisiae. Saccharomyces cerevisiae is a unicellular eukaryote that has become an important tool in microbial genetic techniques. It undergoes meiosis and sporulation which takes place in a single cell. This medium stimulates the sporulation of diploid yeast cells which occurs only when cells are deprived of any carbon source. Potassium acetate enhances the sporulation of diploid strains. This medium contains agar as the solidifying agent.
Quality Control
Appearance of Powder : Cream to yellow coloured, homogeneous, free flowing powder.
Gelling: Firm, comparable with 1.5% Agar gel.
Colour and Clarity : Light yellow coloured, clear to slightly opalescent gel forms in Petri plates.
Cultural Response : Cultural characteristics observed after an incubation at 35-37°C for 18 - 48 hours.
