Denhardt's Solution (100X) is composed of blocking agents and is named after David T. Denhardt who formulated it in 1966. This solution is used in membrane-based nucleic acid hybridization protocols like Southern and Northern blotting. It has been proved that preincubation of nucleic acid containing filters in this solution prevents nonspecific binding of single-stranded DNA probes as the high-molecular weight polymers saturate the nonspecific binding sites and artificially increases the concentration of available probe.
